Bupropion- Stimulates the pro-opiomelanocortin (POMC) neurons and increases production and release of α-MSH and β-endorphin. α–MSH mediates the anorectic effect of POMC which leads to decreased food intake, increased energy expenditure and weight loss. Β-endorphin binds to mu opioid receptors (MOP-R) that are responsible for autoinhibitory feedback, thereby inactivating the anorectic effect, and acting like a brake to reduce activity of POMC cells (see naltrexone).
Naltrexone- increases the activity of pro-opiomelanocortin (POMC) cells by blocking the MOP-R and preventing B-endorphin-mediated autoinhibitory feedback of POMC cells, thereby maintaining an anorectic environment. The combination of bupropion and naltrexone produces a greater increase in POMC activity than either drug alone. Additionally, this combination of bupropion and naltrexone may also regulate the mesolimbic reward pathways, which may lead to further weight reduction by modulating reward values and goal-oriented behaviors.
Topiramate- While the mechanism by which topiramate alters energy balance and induces fat loss is still under investigation, it is thought to be related to topiramate’s inhibitory action on glutamate neurotransmission and neuropeptide Y, as these biomolecules are related to appetite stimulation.
Chromium picolinate- This is a well absorbed form of Chromium, which has a well-documented role in insulin action and glucose metabolism. Studies have shown effects ranging from modulating blood sugar levels to decreasing appetite, hunger cravings, and binge-eating episodes.
